CommentsLipoxygenase is an undesirable enzyme in barley and malt that affects the flavor stability of beer, due to the enzymatic oxidation of linoleic acid to trans-2-nonenal by the LOX pathway. We used a high-throughput screening method for LOX that utilizes ferrous oxidation-xylenol orange (FOX reagent) for the determination of lipid hydroperoxides (Li, 2008). Oxidation of ferrous to ferric ions by lipid hydroperoxides occurs under acidic condition and then Xylenol orange (XO) binds ferric ions to form a chromophore complex, which gives a maximum absorbance at 560 nm. During the LOX assay, in the first step, we add 110 uL phosphate buffer to each tube and add another 30 uL substrate (linoleic acid) to each tube in step 2. Step 3 is an enzymetic reaction step. Starting from the first tube, we add 10 uL enzyme extract to each tube every 30 s. For 10 glass tubes, this process takes 4 min 30 s. Step 4 can be regarded as a step for measuring lipid hydroperoxides content. At the 5th min, we quickly add another 2.85 ml of FOX reagent to the first tube and add same amount of FOX reagent every 30 s to the rest of 9 tubes. At the end of this step, your timer should be 9 min 30 s. In the final step (step 5), we read OD value at 560 nm against reaction blank. Starting at the 15th min, because FOX reaction needs to take 10 min, so we read OD value of each tube by spectrometer every 30 s.
